چکیده

A successful unified pharmacophore/receptor model which has guided the synthesis of subtype selective compounds is reviewed in light of recent developments both in ligand synthesis and structural studies of the binding site itself. The evaluation of experimental data in combination with a comparative model of the 1 2 2 GABAA receptor leads to an orientation of the pharmacophore model within the Bz BS. Results not only are important for the rational design of selective ligands, but also for the identification and evaluation of possible roles which specific residues may have within the benzodiazepine binding pocket.

A Review of the Updated Pharmacophore for the Alpha 5 GABA(A) Benzodiazepine Receptor Model

An updated model of the GABA(A) benzodiazepine receptor pharmacophore of the α5-BzR/GABA(A) subtype has been constructed prompted by the synthesis of subtype selective ligands in light of the recent developments in both ligand synthesis, behavioral studies, and molecular modeling studies of the binding site itself. A number of BzR/GABA(A) α5 subtype selective compounds were synthesized, notably...

Residues at Positions 206 and 209 of the a1 Subunit of g- Aminobutyric AcidA Receptors Influence Affinities for Benzodiazepine Binding Site Ligands

Ligands of the benzodiazepine binding site allosterically modulate g-aminobutyric acidA receptors. Their binding pocket is made up of amino acid residues located on both a and g subunits. We transiently expressed wild-type a1b2g2 and mutant GABAA receptors in human embryonic kidney 293 cells and determined their binding properties. Receptors containing the mutant aY209A showed ;40-fold decrease...

Down-Regulation of Benzodiazepine Binding to a5 Subunit- Containing g-Aminobutyric AcidA Receptors in Tolerant Rat Brain Indicates Particular Involvement of the Hippocampal CA1 Region

Chronic benzodiazepine treatment can produce tolerance and changes in g-aminobutyric acid (GABA)A receptors. To study the effect of treatment on a selected population of receptors, assays were performed using [H]RY-80, which is selective for GABAA receptors with an a5 subunit. Rats were given a flurazepam treatment known to produce tolerance and down-regulation of benzodiazepine binding, or a d...
